UASt regulatory sequences drive expression of an inverted repeat.
The expression of barcGD9921 under the control of Scer\GAL4insc-Mz1407 leads to a significant increase in the number of intermediate neural progenitors, namely of immature Ase-positive, Dpn-negative intermediates, and to significant decreases in the numbers of ganglion mother cells and of Pros-positive neurons in type II neuroblast lineages, as compared to controls. This expression also leads to significant increases in the cell size of type I and type II neuroblasts, intermediate neural progenitors, and ganglion mother cells, as compared to controls. Expression under the control of either Scer\GAL4ase.neuro or Scer\GAL4insc-Mz1407 leads to a significant decrease in the number of neuroblasts and to the converse significant increase in the number of ganglion mother cells in type I neuroblast lineages, as compared to controls.
Individuals expressing barcGD9921 under the control of Scer\GAL4pnt-14-94 are viable and reach adulthood. This expression leads to adult brains exhibiting severe neuroanatomical defects in the central complex (the ellipsoid body, fan-shaped body, and nodoli are all deformed) and exhibiting a decrease in the number of type II neuroblast-derived neurons, as compared to controls.
The expression of barcGD9921 leads to a significant decrease in the division rates of type II neuroblasts and intermediate neural progenitors (under the combined control of Scer\GAL4wor.PA, Scer\GAL80ase.PN) and of type I neuroblasts and ganglion mother cells (under the control of Scer\GAL4ase.neuro alone), as shown by their shorter cell cycle time and their higher proportion of cells in G2/M compared to controls.
Expression under the control of Scer\GAL41407 results in an increase in the number of neuroblasts in the larval brain compared to controls.
Expression under the control of Scer\GAL41407 results in longer neuroblast lineages (more daughter cells per neuroblast) in the larval brain compared to controls.
Expression under the control of Scer\GAL41407 results in mild defects in the shape of ganglion mother cells in the larval brain.
Expression under the control of Scer\GAL41407 results in an increase in the average size of ganglion mother cells in the larval brain compared to controls.
Adults expressing barcGD9921 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
Expression under the control of Scer\GAL4Mef2.PR results in adult/pharate lethality.
Expression under the control of Scer\GAL4pnr-MD237 results in the absence of 100% of the Scer\GAL4pnr-MD237-expressing area of the notum.
