The expression of Cdc16GD12028 under the control of Scer\GAL4ey.PU leads to eye defects, including a mild decrease in eye size, severe eye morphology and polarity defects and the frequent formation of ectopic structures within the eye field that resemble naked head cuticles or antennae (indicative of a complete loss or a conversion of cell fate), as compared to controls; in combination with Dicer-2 (for more efficient RNAi) there is a strong reduction in eye size, as compared to controls. In the third instar larval eye disc, this expression strongly inhibits morphogenetic furrow progression (particularly evident in the lateral margin regions) and retinal differentiation, and leads to a significant increase in mitotic index, without affecting the disc size, as compared to controls; when expressed at 18[o]C temperature, the morphogenetic furrow progression is inhibited in the lateral margins without affecting the mitotic index, as compared to controls.

The expression of Cdc16GD12028 under the control of Scer\GAL4dac-GAL4 leads to morphogenetic furrow progression defects and to a small but significant reduction of the differentiated area in the third instar larval eye disc, as compared to controls. Expression under the control of Scer\GAL4h-1J3 leads to a delay in morphogenetic furrow progression in the lateral regions of the eye disc only when in combination with Dicer-2 (for more efficient RNAi). Expression under the control of Scer\GAL4GMR.PU leads to a rough eye phenotype only under high temperature conditions (29[o]C) or when in combination with Dicer-2 (for more efficient RNAi), as compared to controls.

Expression under the control of Scer\GAL41407 results in shorter neuroblast lineages (less daughter cells per neuroblast) in the larval brain compared to controls.

Expression under the control of Scer\GAL41407 results in loss of neuroblasts in the larval brain associated with either increased or decreased size of the remaining neuroblasts (depending on the insertion line used).

Expression under the control of Scer\GAL41407 may result in mild defects in the shape of neuroblasts in the larval brain (depending on the insertion line used).

Adults expressing Cdc16GD12028 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression under the control of Scer\GAL4pnr-MD237 results in loss of bristles on the notum in 60-70% of the Scer\GAL4pnr-MD237 expression domain.

Expression under the control of Scer\GAL4pnr-MD237 results in the absence of 70-80% of the Scer\GAL4pnr-MD237-expressing area of the notum.

Df(3L)H99 enhances the small eyes induced by the expression of Cdc16GD12028 under the control of Scer\GAL4ey.PU.

The adult eye defects (decreased eye size, and morphology and polarity defects) and most third instar larval eye disc defects (i.e. the morphogenetic furrow progression and retinal differentiation defects, but not the increased mitotic index) induced by the expression of Cdc16GD12028 under the control of Scer\GAL4ey.PU are fully suppressed by the co-expression of either Nek2KK100496 or Nek2HM05088. Conversely, the adult eye morphology defects and the third instar larval eye disc morphogenetic furrow progression and retinal differentiation defects are enhanced by the co-expression of Nek2Scer\UAS.ORF, leading to a severe expansion of the head case; the adult eye morphology defects are also enhanced by the co-expression of either Nek2UAS.cMa.GFP or Nek2Ubi-p63E.GFP.

The third instar larval eye disc morphogenetic furrow progression and retinal differentiation impairment induced by the expression of Cdc16GD12028 under the control of Scer\GAL4dac-GAL4 is suppressed by the co-expression of Nek2KK100496.

The co-expression of Cdc16GD12028 and Nek2Scer\UAS.ORF under the control of Scer\GAL4dpp.PU leads to a delay in third instar larval eye disc morphogenetic furrow progression, as compared to controls.

The co-expression of Cdc16GD12028 does not suppresses the small eyes induced by the expression of CycAGD8653 under the control of Scer\GAL4ey.PU.

Xenogenetic Interactions
Statement
Reference

The retinal differentiation inhibition, but not the increased mitotic index, resulting from the expression of Cdc16GD12028 under the control of Scer\GAL4ey.PU is enhanced by the co-expression of BacA\p35Scer\UAS.cHa, resulting in undifferentiated masses in the adult eye field.

Complementation and Rescue Data
Comments

The third instar larval eye disc differentiation impairment and adult eye morphology defects induced by the expression of Cdc16GD12028 under the control of Scer\GAL4ey.PU are rescued by the co-expression of Cdc16Ubi-p63E.T:Avic\GFP-m6.
