Mobilisation of an X-linked P{GawB} generates this line.
ScerGAL49-20 drives expression in all eight chordotonal organs of the PNS and in multiple medial and lateral cells of the ventral nerve cord. Labelling is also detected in a few cells in the brain.
Inhibition (using Ctet\tetXTNT.UAS) of a subset of Scer\GAL49-20 cells (expression restricted using Scer\GAL80iav.PF) in larvae results in a significant reduction in locomotion speed, a significant increase in peristaltic wave duration, and reduced frequency of forward peristaltic waves, compared to controls; when the number of inhibited cells is reduced by addition of Scer\GAL80tsh-GAL80, there is no difference in peristaltic wave duration or frequency, compared to controls.
Optogenetic activation (using Crei\ChR2T159C.UAS.YFP with blue light) of a subset of Scer\GAL49-20 cells (expression restricted using Scer\GAL80iav.PF) in larvae causes them to stop crawling; larval abdominal, but not thoracic, segments are paralyzed, unlike controls; neuromere-specific activation results in segment-specific body wall muscle relaxation, which can disrupt peristaltic wave propagation; when the number of inhibited cells is reduced by addition of Scer\GAL80tsh-GAL80, there is no difference in larval locomotion, compared to controls.
Thermogenetic activation (using TrpA1UAS.(B).cKa at 32[o]C) of a subset of Scer\GAL49-20 cells (expression restricted using Scer\GAL80iav.PF) in larvae results in a significant reduction in locomotion speed and significantly increased peristaltic wave duration, compared to controls.
