Acute inhibition (using shi1.UAS at 30[o]C) of a subset of Scer\GAL4GMR49H02 cells (expression restricted using Scer\GAL80Mef2.mb247) in adults significantly reduces electric shock avoidance, at a range of voltages, and avoidance of higher temperatures, compared to controls, with no effect on locomotion activity index or odor avoidance; inhibition of this subset during testing alone, but not training alone, significantly reduces short-term aversive, but not appetitive, olfactory memory, compared to controls.
Inhibition (using Hsap\KCNJ2UAS.EGFP) of Scer\GAL4GMR49H02 cells, in combination with a Gal80[ts] transgene to begin expression three days prior to experiments, in adults significantly reduces electric shock avoidance, compared to controls, with no effect on locomotion activity index.
Optogenetic activation (using Crei\ChR1::Vcar\ChR1::Vcar\ChR2ReaChR.UAS.Citrine with (orange) light) of Scer\GAL4GMR49H02 cells in one chamber of a T-maze results in a significant increase in avoidance of that chamber, compared to controls, with no effect on locomotion activity index.
Thermogenetic activation (using TrpA1UAS.(B).cKa at 26[o]C) of a subset of Scer\GAL4GMR49H02 cells (expression restricted using Scer\GAL80Mef2.mb247) in adults, in combination with a paired odor, results in a significant increase in aversive olfactory memory, compared to controls, with no effect on locomotion activity index or odor avoidance.
