UAS regulatory sequences drive expression of a hairpin repeat consisting of cDNA and genomic DNA derived sequences, where the intron in the genomic DNA is spliced out during processing to generate a hairpin loop.
Expression of garzdsRNA.Scer\UAS under the control of Scer\GAL4Hml.Δ results in an increase in third instar larval secondary lymph gland lobe number. Phenotypes are also seen in the primary lobe; an increased number of mitotically active cells is seen in the cortical zone compared to controls and the number of niche (posterior signalling center) cells in the primary lobe is highly reduced compared to controls. Premature differentiation of prohemocytes into plasmatocytes and crystal cells is seen.
Expression of garzdsRNA.Scer\UAS under the control of Scer\GAL4Hml.Δ results in endocytic trafficking defects during larval haematopoesis. NICD is trapped in Hrs positive endosomes.
