Expression of sesBdsRNA.Scer\UAS in Malpighian tubules under the control of Scer\GAL4c42 results in mitochondria becoming distributed throughout the cell, with short and globular morphology, as oppose to being positioned adjacent to the basolateral membrane as in controls. In particular, the normally prominent threadlike brush border is reduced and its mitochondrial density lower.
Targeted expression of sesBdsRNA.Scer\UAS in the Malpighian tubules under the control of Scer\GAL4c42 results in a significantly increased basal cytoplasmic Ca[2+] concentration and abolishes the Capa-1-induced calcium response. Similarly, sesBdsRNA.Scer\UAS expression increases basal Ca[2+][[mit]] levels and a markedly inhibited mitochondrial calcium response to Capa-1.
Targeted expression of sesBdsRNA.Scer\UAS in tubule stellate cells, under the control of Scer\GAL4c710 results in increased basal cytoplasmic Ca[2+] concentration levels and in a significantly decreased Lk-induced calcium response.
Targeted expression of sesBdsRNA.Scer\UAS in tubule principal cells using the Scer\GAL4Uro.PT driver significantly decreases the availability of ATP to the cell and decreases survival upon oxidative stress challenge by H[[2]]O[[2]].
Expression of sesBdsRNA.Scer\UAS in Malpighian tubules under the control of Scer\GAL4c42 results in a reduced rate of fluid transport compared to controls.
Targeted expression of sesBdsRNA.Scer\UAS in tubule principal cells under the control of Scer\GAL4hs.PB results in an increase in mitochondrial activation throughout the cell.
Expression of sesBdsRNA.Scer\UAS in Malpighian tubules under the control of Scer\GAL4c42 or Scer\GAL4hs.PB results in an increase in H[[2]]0[[2]] production both in the tubules and in mitochondria isolated from the tubules.
