Upf214J contains a 14bp deletion (1682-1695) approximately halfway through the 1,241-residue coding sequence of Upf2, which causes a frame shift at codon 562 followed by a stop 40 codon later.

28.9% of eggs derived from homozygous female germline clones show a fused dorsal appendage phenotype. Eggs also occasionally show a "curled" phenotype, in which the ends of the dorsal appendages curl around to form a hook-like shape. None of the eggs hatch.

Homozygous follicle cell clones are smaller than wild-type clones both in early, proliferating egg chambers and in late, endocycling egg chambers.

Mutant third instar larvae can live for up to 13 days before dying. Approximately 10% of these larvae attempt to pupate, but normal head inversion fails to occur.

Upf214J mutants do not exhibit defects in larval cell differentiation. Larval tracheal cells lacking Upf2 display wild-type morphology at all levels of branching, including tracheal terminal cell clones, which show normal branching patterns and luminal structures. Likewise, type IV da neurons appear normal in these mutants.

While homozygous mutant Upf214J clones are readily obtained from the larval tracheal system, clones are not recovered in the adult system. Large clones are also not recovered from the adult epidermis or eyes, Although small peripheral eye clones are occasionally observed.

A WGMR.PG background, used to eliminate all heterozygous and wild-type cells in the developing eye, Upf214J cells proliferate, differentiate, and form an eye, although the eyes are smaller and more disorganised (rougher) than those of controls.
