ScerGAL4Piezo.1.0 drives expression in sensory neurons in the adult and larva. In the adult, these include Type I (eo type) ciliated sensory neurons such as the ones that form the campaniform sensilla of the haltere, the thoracic bristles and the chordotonal neurons from the Johnston organ. In the larva, ScerGAL4Piezo.1.0 labels multidendritic neurons (Type II) including neurons of the class III and the lateral bipolar md neuron (abdominal intersegmental bidendritic neuron isbp). Some of the Type II neurons identified by ScerGAL4Piezo.1.0 are also labeled with ScerGAL4ppk.PG. Expression of ScerGAL4Piezo.1.0 is also detected in larval non-sensory tissues such as the hindgut, salivary glands, aorta and trachea.
crop, with TrpA1UAS.(B).cKa
The acute inhibition of Piezo-positive neurons, by expressing shi1.UAS under the control of Scer\GAL4Piezo.1.0 and testing adults at the non-permissive 32[o]C) increases feeding (also leading to gut bloating and increased crop size). Conversely, acute activation of Piezo-positive neurons, by expressing TrpA1UAS.(B).cKa under the control of Scer\GAL4Piezo.1.0 and activating it 30[o]C, suppresses feeding (and crop size); the same decrease in feeding occurs upon acute activation of a subset of Piezo neurons (expression of TrpA1UAS.(B).cKa under the combined control of Scer\GAL4Piezo.1.0 and Scer\GAL80ChAT.7.4P).
Scer\GAL4Piezo.1.0/TrpA1UAS.(B).cKa is a suppressor of increased rate of feeding behavior | recessive | adult stage phenotype of PiezoKO
Scer\GAL4Piezo.1.0/TrpA1UAS.(B).cKa is a suppressor of increased size | recessive | adult stage phenotype of PiezoKO
Scer\GAL4Piezo.1.0/TrpA1UAS.(B).cKa is a suppressor of crop phenotype of PiezoKO
Acute activation of Piezo-positive neurons, by expressing TrpA1UAS.(B).cKa under the control of Scer\GAL4Piezo.1.0 and activating it 30[o]C, suppresses the increased feeding and crop distention observed in PiezoKO adults.
