UAS regulatory sequences drive expression of the first 4086 nucleotides of the Hsap\KMT2A cDNA fused to nucleotides 1432-1704 of the Hsap\MLLT3 cDNA. The fusion protein is tagged at the C-terminal end with Tag:FLAG.

      The brains of larvae expressing Hsap\MLL::Hsap\MLLT3Scer\UAS.C.T:Zzzz\FLAG under the control of Scer\GAL4da.G32 show increased BrdU incorporation compared to wild type. Analysis of BrdU labelling indicates that the cells show faster mitotic entry than normal. Chromatin structure appears normal in 55% of metaphase spreads in brain squashes of these larvae, but a significant number of mitotic figures are characterised by decondensed portions along the chromosome arms and chromosome fragmentation is seen.
