UASp regulatory sequences drive expression of the CFP form of Avic\GFP, a linker sequence VIETDGA, the YFP form of Avic\GFP and the mitochondrial outer membrane targeting sequence from CG9393.
The linker between the CFP and YFP coding regions contains a IETD cleavage site that is recognised by the Dredd and Nc caspases. Upon cleavage by caspase, the CFP portion is no longer tethered to the mitochondrion-localised YFP and the YFP emission of ~530nm (mitochondrial) and CFP emission of ~480nm (cytoplasm) become spatially distinct.
