Description

FLP-mediated recombination has deleted the genomic sequence between the two progenitor insertions, removing sequence extending from upstream of the second ATG codon of rg through the entire coding sequence and 3'UTR.

Comment:

FLP-mediated recombination has deleted the genomic sequence between the two progenitor insertions, removing most of the coding sequence and all of the 3'UTR.

Detailed Description

Mutants have a rough eye phenotype.

Mutant flies show a complete loss in aversive and appetitive memory when assayed immediately after olfactory conditioning.

68% of mushroom body hemispheres have morphological defects in hemizygous males and 77% of hemispheres have morphological defects in homozygous females. 56% of mushroom body hemispheres have morphological defects in rgFDD/Df(1)ovo41 females.

Hemizygous male, homozygous female and rgFDD/Df(1)ovo41 female third instar larvae have increased bouton numbers per muscle area at the neuromuscular junction compared to wild type.

Quantal content at the neuromuscular junction of mutant larvae is similar to wild type.

Expression of Mmus\NbeaScer\UAS.cVa under the control of Scer\GAL4GMR.PU rescues the rough eye phenotype of rgFDD flies.

The mushroom body defects seen in hemizygous rgFDD males are not rescued by expression of Mmus\NbeaScer\UAS.cVa under the control of either Scer\GAL4elav.PU, Scer\GAL4ey-OK107 or Scer\GAL4Mef2.247.

Expression of Mmus\NbeaScer\UAS.cVa under the control of Scer\GAL4n-syb.PS rescues the increased bouton number at the neuromuscular junction which is seen in hemizygous rgFDD male third instar larvae.

Comments

Expression of rgScer\UAS.cVa under the control of Scer\GAL4GMR.PU rescues the rough eye phenotype of rgFDD flies.

Expression of rgScer\UAS.cVa under the control of Scer\GAL4elav.PU completely rescues the loss of immediate aversive memory seen in rgFDD flies and partially rescues the loss of immediate appetitive memory seen in rgFDD flies in an olfactory conditioning assay.

The mushroom body defects seen in hemizygous rgFDD males are partially rescued by expression of rgScer\UAS.cVa under the control of either Scer\GAL4elav.PU or Scer\GAL4ey-OK107, but are not rescued by expression under the control of Scer\GAL4Mef2.247.

Expression of rgScer\UAS.cVa under the control of Scer\GAL4n-syb.PS rescues the increased bouton number at the neuromuscular junction which is seen in hemizygous rgFDD male third instar larvae.
