Description

141bp spanning the mir-1000 hairpin has been deleted. A single loxP site is present at the site of the deletion. Expression of the msi gene (mir-1000 is located within an intron of msi) is unaffected.

The mir-1000 hairpin has been deleted. A single loxP site is present at the site of the deletion. The expression of the msi host gene appears unaffected (assayed by quantitative PCR).

mir-1000KO1/mir-1000KO2 and mir-1000KO2/Df(3R)Exel6203 adults show a sharply reduced lifespan, with survival declining rapidly by 2 weeks of age. mir-1000KO1/mir-1000KO2 adults show a progressive decline in climbing ability, with significant defects already seen at 2 days of age. Caspase-3 positive cells are seen in the brains of 2 and 10 day old mir-1000KO1/mir-1000KO2 adults and vacuoles are seen in 10 day old brains.

mir-1000KO1/mir-1000KO2 adults show reduced survival on exposure to hydrogen peroxide compared to controls.

The number and size of synaptic boutons at the neuromuscular junction is increased compared to controls in mir-1000KO1/mir-1000KO2-GAL4 larvae.

Homozygous larvae show a significant increase in the amplitude of spontaneous excitatory junction potentials at the neuromuscular junction.

mir-1000KO/Df(3R)Exel6203 males and females show a significant decrease in median lifespan compared to controls.

Expression of Diap1Scer\UAS.cUa under the control of Scer\GAL4mGluR-NP6701 partially suppresses the short lived phenotype of mir-1000KO1/mir-1000KO2 adults and reduces the number of Caspase-3 positive cells seen in the adult brain of these flies. The climbing defects are also rescued.

The climbing defects of two day old mir-1000KO1/mir-1000KO2 adults are rescued if they are also carrying Df(2L)BSC37/+. The amount of Caspase-3 positive cells in the brain is reduced, and lifespan is increased (although not to control levels).
