FlyBase curator comment: this construct is based on the Super-Clomeleon construct described in Grimley et al., 2013 (PMID: 24107961). The protein consists of two different chromophores (the Topaz YFP and Cerulean variants of Avic\GFP) separated by a short linker sequence. The YFP contains a Cl[-] binding site adjacent to its chromophore and Cl[-] binding results in a conformational change that can be detected as a change in the efficacy of fluorescence resonance energy transfer (FRET) between the two Avic\GFP variants. YFP contains mutations (Q69T and V163A) that optimise Cl[-]binding, and both Cerulean and YFP contain a mutation (S30R) that increases brightness and stability.
