20xUAS regulatory sequences drive expression of a mutated form of lin-28 (carries mutations in the CCHC domain that converts amino acids at positions 136 and 158 to Ala residues) which is tagged at the C-terminal end with three copies of Tag:FLAG and then three copies of Tag:HA.

      Expression of lin-28M2.20xScer\UAS.T:Zzzz\FLAG,T:Ivir\HA1 under the control of Scer\GAL4esg.PU in adult intestinal stem cells (ISCs) and enteroblasts for two days (using the temperature sensitive Scer\GAL80ts.αTub84B line to control the timing of expression) results in a significant increase in the total number of ISCs in the posterior midgut compared to in controls.
