UASt regulatory sequences drive expression of a full-length spidey open reading frame (ORF) that is tagged at the C-terminal end with three copies of the Tag:HA epitope. The ORF is flanked by a pair of incompatible FRT sites (FRT5 and FRT2), which allows for future in vivo exchange of either the promoter or the C-terminal tag sequence.
Expression of spideyScer\UAS.ORF.GW.T:Ivir\HA1 under the control of Scer\GAL4Desat1.PB with Scer\GAL80ts.αTub84B results in complete lethality either during prepupal stage or pharate adult stage when the flies are reared under restrictive temperature (29[o]C) and partial lethality when kept under permissive temperature (19[o]C). spideyScer\UAS.ORF.GW.T:Ivir\HA1 expressing flies also display head and spiracle eversion defects.
Expression of spideyScer\UAS.ORF.GW.T:Ivir\HA1 under the control of Scer\GAL4Desat1.PB with Scer\GAL80ts.αTub84B leads to a significant drop in the levels of steroid hormones 20-hydroxyecdysone and makisteron A (while the amounts of their catabolic products are below detection level) in third instar larvae when the animals are reared under restrictive temperature (29[o]C) compared to controls kept under permissive temperature (19[o]C).
