Ten copies of the UAS regulatory sequence drive expression of a light-activated caspase which is tagged at the C-terminal end with mCherry. The light-activated caspase consists of a non-cleavable form of Hsap\CASP3 with a light-sensitive Tag:AsLOV2 domain inserted into the intersubunit linker. The normal mechanism of Hsap\CASP3 activation by cleavage of the intersubunit linker (followed by dimerization of the two released subunits) has been abolished by introducing the mutations D174N and DDD(179-181)NNN. In its place, a light-dependent conformational change of the Tag:AsLOV2 domain allows this non-cleavable form to fold into an active caspase. The Tag:AsLOV2 domain contains two mutations, G528A and N538E, to increase the dark stability (PMID:25988185). Seven Ala residues have been added to the linker N-terminal to the Tag:AsLOV2 tag, and seven residues of the intersubunit linker have been deleted from the C-terminal side.
