UAS regulatory sequences drives expression of a constitutively active form of sqh. The protein is tagged at the N-terminal end with the Tag:ALD(insc) apical localization domain and at the C-terminal end with mCherry.
Expression of sqhEE.Scer\UAS.ALD.T:Disc\RFP-mCherry under the control of Scer\GAL4wor.PA (thus forcing constitutively activated Myosin to the apical cortex) in third instar larval neuroblasts leads to abnormalities in cell shape changes from early anaphase to telophase during asymmetric cell division: the furrowing is initiated closer to the apical surface than in wild-type neuroblast (and no subsequent basal shifting of the ingression is observed). Occasionally, the sqhEE.Scer\UAS.ALD.T:Disc\RFP-mCherry expression can cause the neuroblasts to divide symmetrically or with inverted physical asymmetry.
