bchs17M/Df(2L)cl7 transheterozygous larvae exhibit a mild decrease in the number of RP2 motor neurons, compared to controls. Their brains exhibit decreased autophagosome number (Atg8 spots/cell) and increased autophagic flux.
Cultured larval neurons exhibit significantly more Atg5-positive vesicles, particularly enriched in the neurites, and significantly fewer Atg8-positive autophagosomes, as compared to controls. Additional expression of bchsUAS.1.GFP under the control of Scer\GAL4eve.PU fully suppresses the fewer autophagosomes and even leads to a severe increase in their number. Both Atg5-positive vesicles and Atg8-positive autophagosomes accumulate in the neurites in differentiated (i.e. 4-day cultured) neurons.
