UASt sequences regulate expression of membrane-tethered form of nkt in a configuration in which the nkt protein is extracellular, with the C-terminal end of nkt being free. The encoded protein consists of the Tag:TM(hTNF) transmembrane domain, a Tag:MYC tag that is flanked on both sides by a GN-repeat linker, and the nkt coding sequence (minus the signal peptide). The protein is predicted behave as a cell-autonomous peptide ligand that acts on the same cell if a cognate receptor is expressed.
