Gene Summary
bloated tubules (blot) encodes a member of the sodium- and chloride-dependent neurotransmitter symporter family. It is required for cortical actin organisation and contributes to embryonic cellularization and renal tubule development in larvae. [Date last reviewed: 2019-03-07] (FlyBase Gene Snapshot)

Gene Group (FlyBase)
SLC6 FAMILY OF SODIUM/CHLORIDE-DEPENDENT NEUROTRANSMITTER TRANSPORTERS -
Solute carrier family 6 (SLC6) members are Na+/Cl--dependent transporters of extracellular neurotransmitters, amino acids and osmolytes across the plasma membrane into the cytosol. (Adapted from FBrf0193583).

Structure
Protein 3D structure   (Predicted by AlphaFold)   (AlphaFold entry Q8IQQ7)

If you don't see a structure in the viewer, refresh your browser.
Model Confidence:
  • Very high (pLDDT > 90)
  • Confident (90 > pLDDT > 70)
  • Low (70 > pLDDT > 50)
  • Very low (pLDDT < 50)

AlphaFold produces a per-residue confidence score (pLDDT) between 0 and 100. Some regions with low pLDDT may be unstructured in isolation.

Testis-specificity index

The testis specificity index was calculated from modENCODE tissue expression data by Vedelek et al., 2018 to indicate the degree of testis enrichment compared to other tissues. Scores range from -2.52 (underrepresented) to 5.2 (very high testis bias).

Additional Descriptive Data

blot transcript is detected at all embryonic stages, with highest expression at 0-3 hr AEL and 3-6 hr AEL. blot transcript is evenly distributed in early embryos. At embryonic stage 5 blot transcript is localized to an anterior cap and in three broad ectodermal stripes. At embryonic stage 6 blot transcript is expressed in five strong and one weak ectodermal stripes, which are interrupted by a ventrolateral region of non-expression. There is also a posterior-lateral stripe of expression at the rim of the amnioproctodeal invagination. By stage 13, most blot transcript expression is limited to the Malpighian tubule tip cell primordium. At stage 16, blot transcript is expressed in a segmentally repeated pattern in the ventral nerve cord, in the antennomaxillary complex, and in the posterior spiracles.

blot is required for viability. Mutants show defects in the Malpighian tubule cells of larvae, and the actin filaments in the apical cortex and the cortical cytoskeleton of embryos, and the cellularisation process.

        Additional comments

Comparison of restriction maps for the genomic regions of tap and blot reveals no overlap. Whole mount in situ hybridization with fragments of the blot walk do not stain tap- expressing cells. tap and blot therefore have distinctive genomic organizations.
