The P{PdL} element contains tetO regulatory sequences and an Hsp70 core promoter at its 3' end, oriented so that transcription from the promoter is directed outwards from this end of the element. This can result in misexpression of neighbouring genomic loci when the element is inserted in the genome. Expression is conditional, depending on the presence of a driver that binds the tetO regulatory sequence, such as the tTA or rtTA driver. In addition, the activity of such drivers can be regulated by feeding with tetracycline, allowing further temporal control of expression.
