Ten copies of the UAS regulatory sequence are separated from a non-fluorescent 'spaghetti monster GFP' ('smGdP') marker by an FRT cassette containing stop sequences, which prevents expression of the marker in the absence of FLP recombinase. The smGdP marker consists of 10 copies of the Tag:HA tag inserted in groups into the backbone of the superfolder green fluorescent protein, which has been rendered non-fluorescent by mutation. A N-terminal myristoylation signal (Tag:Myr(Src64B)) is also present and the marker has been optimised for Drosophila codon usage.
Ten copes of the UAS regulatory sequence drive expression of superfolder GFP which is tagged with a myristoylation signal (Tag:Myr(Unk)).
Tandem vector containing two open reading frames (separated by a 2.8kb gypsy-insulated spacer): a superfolder GFP marker and a non-fluorescent 'spaghetti monster GFP' ('smGdP') marker (Avic\GFP10xScer\UAS.Scer\FRT.smGdP.T:Myr-Src64B,T:Ivir\HA1) tagged with T:Ivir\HA1. The smGdP marker is separated from its regulatory sequence by a Scer\FRT cassette containing stop sequences, preventing its expression in the absence of FLPase.
