Comment:

frameshift after this amino acid (993) results in the substitution of 30 new residues in place of the last 9 amino acids; exact location/nature of mutation unspecified.

Detailed Description

dor4 mutant males exhibit ~60% less spermatogonial cyst death compared to controls.

dor4 male wandering third instar larvae show defects in morphology of LysoTracker-positive puncta and lipid droplets during ecdysone-induced autophagy.

92% of dor4 stage 14 egg chambers show persisting nurse cell nuclei, compared to 7% in controls.

Mutant males have a shortened adult life span compared to controls, with a mean life span of 13.94 days at 25o.

Hemocyte endosomal degradation is significantly reduced in dor4 mutants compared to wild-type cells. This reduction also correlates with an increased severity of eye-colour defect. This reduction also correlates with an increased severity of eye-colour defect (with increased severity in dor1 compared to dor4. Distinct from cells in wild-type and car1 animals, the large endosomes in mutant dor4 alleles appear blocked in progression to later stages.

Synapse size, along with bouton number, is decreased in dor4 mutants.

Eye colour: orange.

Eye colour: dark orange at 25oC.

Genetic Interactions
Statement

Less than 5% of dor4 hk11 double mutants eclose into adults, with the lethal phase occurring in the late pupal stage. dor4 hk11 double mutants have a decreased number of boutons and are indistinguishable from dor4 mutants alone. However, in the presence of a dor duplication, such as Dp(1;Y)1E, the size of the dor4 hk11 double mutant synapse is like hk11 mutants.

Complementation and Rescue Data
Comments

The defect in degradation in the dor4 mutant cells is completely rescued by Dp(1;Y)1E.

The dor4/dor8 transheterozygote exhibits the same phenotype as the dor4 homozygote.

The eye colour phenotype of dor4 flies is rescued by dor1-1002.Scer\UAS or dor400-1002.Scer\UAS expressed under the control of Scer\GAL4GMR.PF. The eye colour phenotype of dor4 flies is not rescued by dorΔ401-800.Scer\UAS or dor1-802.Scer\UAS expressed under the control of Scer\GAL4GMR.PF, and the level of eye pigmentation in these flies is further reduced compared to dor4 single mutant flies.
