spn-D1 females lay eggs which show patterning defects, including having two dorsal appendages that are fused at the base, having a single dorsal appendage, and having no dorsal appendages, being completely ventralised.

spn-D1 mutants have a similar frequency of single-strand annealing repair (SSA) compared to controls in a P{wIw.FRT} hemizygous assay to study DNA double-stranded break repair when assayed at 38oC.

spn-D2/spn-D1 larvae are not sensitive to MMS.

spn-D2/spn-D1 females exhibit low fertility, and meioses show reduced frequency of crossing over along most of chromosome 2, as compared to wild type.

78% of eggs derived from homozygous females have mutant eggshells, 22% have wild-type eggshells. 70% of eggs derived from spn-D1/spn-D2 females have mutant eggshells and defects in karyosome morphology are seen in the mutant egg chambers. spn-D1 animals show the same survival rate after exposure to X rays or to 0.08% methyl methanesulfonate as control animals.

Homozygous and hemizygous females produce eggs with a range of eggshell phenotypes; from resembling wild-type with two normal dorsal appendages, having two dorsal appendages fused at the base, having a single dorsal appendage to having little or no dorsal appendage material. Hemizygotes are viable. spn-D1/spn-D2 larvae show normal sensitivity to methyl methanesulfonate.

weak maternal-effect lethal. Egg shape affected; in extreme cases dorsal appendages are lacking and eggs have little or no dorsal-ventral polarity; some eggs have one fused dorsal appendage. Low fecundity; eggs often slightly collapsed. embryos sometimes hatch.

The eggshell patterning defects of eggs laid by spn-D1 females are strongly suppressed by Brca2KO/Brca256E.

The eggshell defects seen in eggs derived from homozygous spn-D1 females are almost completely suppressed if the females are also homozygous for mei-W68unspecified; 98% of the eggshells are wild-type in eggs derived from double mutant females. The eggshell defects seen in eggs derived from spn-D1/spn-D2 females are completely suppressed if the females are also homozygous for mei-W68unspecified. The karyosome defects seen in spn-D1/spn-D2 egg chambers are also suppressed by mei-W68unspecified. The eggshell defects seen in eggs derived from homozygous spn-D1 or spn-D1/spn-D2 females are suppressed if the females also carry mei-41unspecified. The karyosome defects seen in spn-D1/spn-D2 egg chambers are only slightly suppressed by mei-41D1 but are strongly suppressed by mei-41D3. spn-D1 spn-BBU double mutant animals show the same survival rate after exposure to X rays or to 0.08% methyl methanesulfonate as control animals.
