Homozygous clones in the pupal wing intermingle with surrounding wild-type cells (in contrast to wild-type clones which have discrete borders). The cells are often dispersed into wild-type tissue in pairs or groups of 4 cells. The mutant cells still respect the lineage restriction at the anterior-posterior boundary. The mutant cells are not the normal hexagonal shape and the area of the apical, but not the basal, surface of the cells is reduced relative to wild-type wing cells. 82% of mutant cells in clones in the pupal wing have adherens junctions condensed into a contact with just one neighbouring cell, in contrast to wild type where there is an even distribution of adherens junction components around the apical circumference of the cell. Clusters of adherens junctions are seen only at interfaces between mutant cells, and not between a mutant and a wild-type cell. Homozygous clones in late third larval instar wing discs are not dispersed, but adherens junctions components are mislocalised, indicating adherens junction mislocalisation precedes dispersal. Dispersal of the mutant cells is first seen 2 hours after pupariation and is initially more pronounced at the distal end of the evaginating wing.

Daily 30 minute heat shocks from day 3 to day 7 of development rescue the larval lethality of Rhs.PA,RrvB1/Df(3L)R-E. Flies are morphologically wild type - except under minimal heat shock conditions when eyes are roughened. Rescued flies can survive without further heat shock for weeks though show a higher mortality than their wild type siblings. Rescued females stop laying eggs within a day or two of heat shock cessation. The ommatidia of partially rescued flies with roughened eyes do not show additional R7 cells. Rhs.PA,RrvB1/Df(3L)R-E mutant females, 7 days after cessation of heat shock, show fragmentation of nurse cell and follicular nuclei. Structure of egg chamber is collapsed with cytoskeleton remains in a single spherical mass. Prior to collapse discontinuities of the follicular epithelium are observed, stalk cells are missing between egg chambers, nurse cells can be reduced in number. Clonal analysis indicates a role for R in the follicular epithelium. Comparing clones induced at various stages reveals a role for R in the early stage of egg chamber development, possibly in the germarium.

Wild type eyes. Transheterozygotes with Df(3L)R-E are late larval lethal.

PDZ-GEFk13720/PDZ-GEF3 females have two spermathecae, but with the introduction of Rap1rvB1/+ or Rap2lSH0581/+ extra spermathecal capsule materials are observed in some females; PDZ-GEFk13720/PDZ-GEF3 wing and eye phenotypes are also enhanced.

Dominantly suppresses the eye phenotype caused by C3GScer\UAS.T:Hsap\MYC,T:Ggal\Myr3 expressed under the control of Scer\GAL4GMR.PF.

Mutation has no effect on the rough eye phenotype caused by two insertions of P{GMR-Rho1}.

Xenogenetic Interactions
Statement
Reference

Twice daily heat shock of Hsap\RAP1Ahs.PA,RrvB1/Df(3L)R-E generates adult flies but the eyes are roughened and rescue is not as robust as with Rhs.PA.

Fail to produce mitotic clones in the eye, this is consistent with an essential role of R gene product in cell proliferation or cell viability.
