Overexpression of wgScer\UAS.cHa by Scer\GAL4C380 results in an increase of ghost boutons upon 5X K[+] spaced depolarisation, similar to controls, but 3X K[+] spaced depolarisation also results in an increase in ghost boutons unlike with controls.

Neuromuscular junctions in which wgScer\UAS.cHa is overexpressed by Scer\GAL4C380 show on average no difference in the mEJP (spontaneous excitatory potentials) frequency when stimulated with 5X K[+] spaced depolarisation, but show a shift toward higher mEJP frequencies when compared in a probability plot.

Expression of wgScer\UAS.cHa under the control of Scer\GAL4dpp.blk1 results in the formation of ectopic pseudotrachea.

When expression is driven by Scer\GAL4arm.T:Hsim\VP16, embryos show puckering of dorsal epidermis with loss of dorsal hair identity and a naked scar at the dorsal midline.

The denticle belts are replaced with an almost uniform expanse of naked cuticle when wgScer\UAS.cHa is expressed under the control of Scer\GAL4e22c.

Expression of wgScer\UAS.cHa under the control of Scer\GAL4prd.RG1 results in the ablation of a portion of the even-numbered denticle belts by excess naked cuticle. Expression of wgScer\UAS.cHa under the control of Scer\GAL4en-e16E results in only subtle changes to the normal denticle pattern and some excess naked cuticle.

Overexpression of wgScer\UAS.cHa in embryos using Scer\GAL4hs.PB between stages 7 and 10 results in a naked cuticle phenotype.

Embryos expressing wgScer\UAS.cHa under the control of Scer\GAL4how-24B have an abnormal second midgut constriction.

Expression of wgScer\UAS.cHa using Scer\GAL4e22c in wgl-17 embryos causes all ventral epidermal cells to secrete naked cuticle. wgl-17 embryos expressing wgScer\UAS.cHa under the control of Scer\GAL4wg.PM show full rescue of the ventral cuticle pattern, although the embryos are slightly distorted as the dorsal pattern elements are not rescued.

Embryos expressing wgScer\UAS.cHa under the control of Scer\GAL4da.G32 in a otk3 background have no denticles.

The phenotype caused when shiK44A.Scer\UAS is expressed at moderate levels under the control of Scer\GAL4prd.RG1 can be rescued by co-expression of wgScer\UAS.cHa; the naked cuticle region in odd-numbered segments is restored to its normal expanse and the even-numbered denticle belts show a more normal degree of diversity. Expression of shiK44A.Scer\UAS under the control of Scer\GAL4en-e16E perturbs denticle type specification in the first three rows of denticles in each segment. The phenotype can be rescued by co-expression of wgScer\UAS.cHa.

Comments

Expression of wgScer\UAS.cHa under the control of Scer\GAL4prd.RG1 in wgl-17 embryos fully rescues naked cuticle in odd-numbered segments and substantially rescues denticle diversity in the epidermal cells adjacent to the Scer\GAL4prd.RG1 expression domain.

wgl-12/+ raised at the restrictive temperature fail to show the increase in ghost boutons seen in controls upon spaced 5X K[+] depolarisation. Expression of wgScer\UAS.cHa using Scer\GAL4C380 in the wgl-12/+ background rescues this phenotype.

The enhancement in numbers of ghost boutons in Scer\GAL4C380/+;;wgScer\UAS.cHa/+ mutants upon 3X K[+] spaced depolarisation is suppressed with wgl-12.
