Stage 13 embryos that are maternally and zygotically mutant for Rbf14 show inappropriate accumulation of BrdU in the epidermis.

Stage 14 embryos that lack maternal and zygotic Rbf14 show an increase in BrdU-positive cells in the anterior and posterior midgut, suggesting that the gap phase of the cell cycle is shorter in these cells.

Rbf14 clones in third instar larvae, result in S phase entry in regions of the zone of nonproliferating cells (ZNC) that are normally arrested in G1.

In Rbf120a/Rbf14 animals, FACS profiles of DNA content in ovarian follicle cells show a population with a ploidy of 32n not seen in wild-type.

Rbf120a/Rbf14 follicle cells show increased ploidy compared to control cells by FACS analysis.

Rbf120a/Rbf14 egg chambers have a mosaic of follicle cells with DNA replication defects. Overamplification of DNA is also seen in the follicle cells. The follicle cells also have cell-cycle defects; the follicle cells undergo at least one extra S phase. DNA replication also persists in later stages of mutant follicle cells than normal.

Some follicle cells from Rbf14/Rbf120a ovaries undergo an additional endocycle.

Homozygous clones in wing discs are not significantly different in size to their wild-type sister clones. Many pyknotic nuclei are associated with mutant clones, suggesting elevated levels of apoptosis. Clones are made in the eye exhibit slight to moderate hypoplasia and mild defects such as missing or duplicated inter-ommatidial bristles and fused ommatidia.

Most homozygotes hatch into larvae and die at an early larval stage. Adult Rbf14 flies rescued by expressing Rbfhs.PD during the first 3 days after egg laying show a number of alterations in adult structures such as abnormal macrochaetae on the notum and rough eyes. All these adult phenotypes are rescued when Rbf function is provided throughout development using heat shock to express Rbfhs.PD.

The eye tumour phenotype of flies expressing DlScer\UAS.cDa under the control of Scer\GAL4ey.PH and also carrying the P{GSV1}lolaGS88A8 insertion (which results in the lolaGS88A8 and psqGS88A8 alleles being expressed under the control of Scer\GAL4ey.PH) is dominantly enhanced by Rbf14.

The cell-autonomous block of cell cycle progression in the position of the second mitotic wave in Df(1)N-54l9 clones in the eye disc is not rescued when clones are also mutant for Rbf14.

Eye disc precluster cells mutant for both dap4 and Rbf14 continue to cycle instead of arresting. In addition, no G1 arrest occurs anterior to the morphogenetic furrow, all cells enter the second mitotic wave, and cells continue to cycle more posteriorly.

Rbf14 does not genetically interact with vriunspecified in double heterozygous combination.

The early larval lethality of hemizygous males is partially suppressed by E2f91/E2f164; these larvae survive to the third larval instar stage. Their development is delayed by about 2 days compared to wild type, and most of the larvae can develop further into pupae. The Rbf14/Y ; E2f91/E2f164 larvae and pupae are slightly smaller than wild type. Some develop into very late stage pupae with adult structures (such as eyes, legs, wings, cuticle and bristles) but they cannot completely eclose. The pattern of BrdU incorporation in E2f91/E2f164 eye discs is the same as wild type. There are two differences in the pattern of BrdU incorporation in the eye discs of Rbf14/Y ; E2f91/E2f164 larvae compared to wild type. Firstly, the G1 arrested region in the morphogenetic furrow is much narrower, cells just anterior to the morphogenetic furrow are not arrested in G1, instead they become part of the first mitotic wave. Secondly, some cells posterior to the second mitotic wave still incorporate BrdU. Rbf14/Y ; E2f91/E2f164 eye discs are slightly smaller than normal. There is increased cell death (seen just anterior to the morphogenetic furrow) in these eye discs compared to those of wild-type or E2f91/E2f164 larvae. The phenotypes of adult Rbf14 flies rescued by expressing Rbfhs.PD during the first 3 days after egg laying can be rescued without heat shock if the flies are also carrying E2f91/E2f164. Approximately 50% of the expected number of Rbf14/Y ; E2fi2/E2frM729 flies survive to the adult stage, and the development of these flies is not significantly delayed compared to sibling flies. The surviving flies have normal eyes, normal macrochaetae on the notum and normal wings and legs. The larval lethality of E2f91/E2frM729 transheterozygotes can be suppressed by Rbf120a/Rbf14; the larvae develop into large late third instar larvae and many can pupate. Some Rbf120a/Rbf14 ; E2f91/E2frM729 animals can develop into pharate adults with adult eyes, legs and wing structures. The development of these larvae is significantly retarded compared to wild-type larvae, with the earliest pupae observed at around day 11 after egg laying. BrdU incorporation is not seen in the optic lobe region of the brain in day 13 E2f91/E2frM729 larvae, in contrast to wild-type third instar larvae. The normal pattern of DNA replication in the optic lobe region is restored if the larvae are also carrying Rbf120a/Rbf14. The eye discs of E2f91/E2frM729 larvae are very small and have only a few cells labelled with BrdU with no specific pattern. In Rbf120a/Rbf14 ; E2f91/E2frM729 eye discs the overall pattern of DNA replication is normal. No significant cell death is seen in these eye discs. In late third instar discs there are about 25 rows of developing photoreceptor cells (as in wild-type discs) but there are much fewer cells anterior to the morphogenetic furrow than in wild type.

Enhances the rough eye phenotype of flies expressing both E2fGMR.PD and DpGMR.PD.
