Expression of GluRIIAM614R.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4Mhc.PW in larval neuromuscular junctions results in decreased amplitude of miniature excitatory junction currents (mEJCs) as compared to controls, EJCs are maintained at wild type levels, and quantal content is increased.

In neuromuscular junctions of GluRIIAM614R.Scer\UAS.T:Hsap\MYC; Scer\GAL4C142 larvae there is a decrease in quantal size, but an increase in quantal content. (Data from electrophysiological recordings made from muscle 6 in segment A3 of wandering 3rd instar larvae).

Expression of two copies of GluRIIAM614R.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4how-24B results in lethality. Expression of one copy of GluRIIAM614R.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4how-24B results in viable adults which do not have any obvious behavioural abnormalities. There is a significant reduction in the quantal size at the neuromuscular junction.

Lrrke03680/+ blocks the increased quantal content seen in larval NMJs expressing GluRIIAM614R.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4Mhc.PW, via a decrease in excitatory junction current (EJC) amplitude.

The increase in quantal content in neuromuscular junctions of GluRIIAM614R.Scer\UAS.T:Hsap\MYC; Scer\GAL4C142 larvae is partially suppressed by CaMKIIT287D.Scer\UAS : There is an decrease of 25% (p < 0.01) (55% decrease when corrected for nonlinear summation) in quantal content compared to GluRIIAM614R.Scer\UAS.T:Hsap\MYC; Scer\GAL4C142 larvae. This decrease is due to a significant reduction (p < 0.01) in the amplitude of excitatory postsynaptic potentials (EPSPs). There is no effect on mini-EPSPs. (Data from electrophysiological recordings made from muscle 6 in segment A3 of wandering 3rd instar larvae). witHA4/witHA5 suppresses the increase in quantal content seen in the neuromuscular junctions of GluRIIAM614R.Scer\UAS.T:Hsap\MYC; Scer\GAL4C142 larvae. (Data from electrophysiological recordings made from muscle 6 in segment A3 of wandering 3rd instar larvae). witHA4/witHA5 suppresses the increase in the excitatory postsynaptic potential amplitude seen in the neuromuscular junctions of Rat\CaMKIINĪ±I.Scer\UAS.cHa; Scer\GAL4C142 larvae. (Data from electrophysiological recordings made from muscle 6 in segment A3 of wandering 3rd instar larvae). The excitatory postsynaptic potential (EPSP) and mini-EPSP amplitudes are significantly decreased in witHA4/witHA5 larvae by the presence of GluRIIAM614R.Scer\UAS.T:Hsap\MYC with Scer\GAL4C142. (Data from electrophysiological recordings made from muscle 6 in segment A3 of wandering 3rd instar larvae).

The addition off GluRIIAM614R.Scer\UAS.T:Hsap\MYC to hiw mutants leads to a an 18% decrease in quantal size, no change in the size of the evoked response, and a compensatory 30% increase in quantal content.
