In dispS037707 larvae, the wing disc pouch is small, the wing disc cells show less abundant and shorter microvilli, as well as fewer extracellular vesicles.

First instar larvae in which dispS037707 germline clones have been induced exhibit only type 4 cuticle cells.

The ventral cuticles of dispS037707 germ-line clone embryos lack naked cuticle, instead forming a lawn of small denticles different from row 5 type.

Homozygotes lacking zygotic disp function survive until early pupal stages. Animals lacking both maternal and zygotic disp function die during embryogenesis with a strong segment-polarity phenotype; the embryos display a lawn of denticle belts and fail to secrete ventral cuticle. The embryos are rescued by a disp+ paternal chromosome. Homozygous clones in the wing can contribute to wild-type wing margin structures. Large clones in the posterior compartment cause a significant reduction in the distance between veins L3 and L4. The wing is reduced in size, but otherwise patterning remains normal.

Expression of hhScer\UAS.cIa, under the control of Scer\GAL4unspecified in dispS037707 mutant embryos rescues some type 1 cuticle, but no type 2 or type 1 cuticle is formed.

Expression of hhScer\UAS.cIa with Scer\GAL4en-e16E in dispS037707 germ-line clone embryos doesn't rescue naked cuticle formation, but does increase denticle diversity in the ventral cuticle: row 3,4 and 5 type denticles are all present. Expression of hhN.Scer\UAS.cGa with Scer\GAL4en-e16E in dispS037707 germ-line clone embryos doesn't rescue naked cuticle formation, but does increase denticle diversity in the ventral cuticle: row 3,4 and 5 type denticles are all present: denticles type 3 and 4 cover each segment, whereas type 5 denticles have almost totally disappeared.

The overgrowth of the anterior compartment of the wing disc seen in larvae expressing hhN.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4en-e16E in a hhts2 background is still seen if the discs are also carrying dispS037707. The ability of hhSF.Scer\UAS.T:Rnor\CD2 to rescue the growth defects of hhts2 wing imaginal discs when expressed under the control of Scer\GAL4en-e16E is unaffected if the discs are also homozygous for dispS037707.

Comments

dispScer\UAS.T:Ivir\HA1 rescues dispS037707 animals to adulthood when expressed under the control of Scer\GAL4en-e16E. The rescued flies have normal patterning in the wing, leg, notum and abdomen and give rise to viable offspring, but have a dramatic reduction in eye size.
