Expressing vvlUAS.cCa under the control of Scer\GAL4c564 results in severe organ malformation or absence in newly eclosed adults.

In vvlScer\UAS.cCa; Scer\GAL4Lim3.B embryos, transverse nerve axons leave the ventral nerve cord normally, but are mistargeted to the ISNb muscle field - either sending collaterals into ISNb muscle targets or, in some cases, innervating these muscles.

When vvlScer\UAS.cCa is driven in the anterodorsal projection neurons (adPNs), the innervation of dorsal landmark glomeruli is largely unaffected, and loss of dendritic projections to these glomeruli are rarely observed. However, these adPNs also mistarget their dendrites to 3 out of 10 lateral landmark glomeruli, DA1 DA2 and VA4. Single cell clones expressing vvlScer\UAS.cCa do not exhibit a detectable phenotype.

Flies expressing vvlScer\UAS.cCa under the control of Scer\GAL4Ser.PGF at 25oC produce wings with occasional blistering within the region of Scer\GAL4Ser.PGF expression at the distal end of the wing. Ectopic growth of intervein tissue is consistently observed resulting in a small outgrowth at the most distal wing tip. The outgrowths do not appear to be organised with the characteristics of a partial wing duplication but seem to result from aberrant cell proliferation at the distal tip of the wing.

vvlScer\UAS.cCa; Scer\GAL4lab.PH partially suppresses the brain phenotype of lab14 homozygous embryos: longitudinal axon pathways in the tritocerebrum are restored but the tritocerebral commissure remains absent and axonal projection of the frontal connective remain abnormal.

The addition of vvlScer\UAS.cCa (driven by Scer\GAL4GH146) to acj66 single cell clones in DL1 leads to a complete failure to innervate the glomerulus DL1. Most of their dendrites partially innervated one or two glomeruli anterior to DL1, with some wandering dendrites within the dorsal half of the antennal lobe. This combination of mutants also leads to a novel stereotyped phenotype in DL1 projection neurons. The dorsal branch has additional side branches; the lateral branch stops well short of the lateral edge of the lateral horn and instead curves and branches to innervate the ventral area of the lateral horn nerver innervated by wild-type DL1 axons.

Expressing vvlUAS.cCa under the control of Scer\GAL4hs.PB together with Avic\GFPS65T.CecA1 is lethal during development, but females can reach adulthood under lower expression conditions (18[o]C).

Comments

Expression of vvlScer\UAS.cCa under the control of Scer\GAL4CG11873-NP2646 partially rescues the Tm27 neuron phenotype of homozygous vvldfr-E82 clones.
