Wings are reduced in size compared to controls in mutant adults. Dorsal bristles in the mutant wing margin are partially ventralised.

Most ash2S112411 homozygotes die as pharate adults. Escapers are sterile, have reduced wing size, a crooked L2 and ectopic vein tissue, mainly extra cross-veins and thicker veins. Late third instar wing discs in these mutants are smaller than wild-type.

About 12% of homozygotes develop into sterile adults that survive for two days. These files show an anomalous pattern of differentiation of the appendages, often associated with homeotic transformations. One example of this is a transformation of the first to the second thoracic segment as observed by reduction of the sex comb teeth, and the occasional partial transformation of the haltere to wing. Mutant wings contain supernumerary cross-veins with 95% penetrance, they are seen in the proximal regions between wing veins L2 and L3, and L4 and L5. Differentiation elements such as longitudinal veins are normally distributed and an overall reduction in wing size is observed. Occasionally other phenotypes are seen: Notches along the posterior margin, winder L3 and L5 wing veins, and distorted L2 veins.

rhove-1 vn1 double homozygotes lack wing veins. This phenotype is partially suppressed by ash2S112411 /ash2S112411. The degree of suppression varies from development of L2 only to almost complete rescue, albeit with some abnormalities such as extra crossvein material or proximal fusions between L2 and L3 or L4 and L5. In 25% of cases, hollow, tube like wings are formed, probably due to detachment of dorsal and ventral layers.

EgfrE3/+ adults have extra vein tissue at the distal end of L2. This phenotype is enhanced by ash2S112411/ash2S112411 - the resulting wings are more reduced and have more ectopic veination than ash2S112411/ash2S112411 animals.

Heterozygous bs03267 flies have small patches of ectopic vein in the vicininty of L2 and L5. If these flies are also homozygous for ash2S112411 their wings are small, blistered and exhibit localised reduction and loss of intervein betwee L2 and L3 and between L4 and L5 and ectopic crossveins. Fusions occur between L2 and L3 and between L4 and L5, particularly proximally.

ash2S112411/ash2S112411 suppresses the loss of L2 seen in kniri-1/kniri-1 flies.

cv-21/cv-21, ash2S112411/+ flies have normal crossveins. cv-21/cv-21, ash2S112411/ash2S112411 have extra crossveins and reduction in wing size.

Comments

The reduced wing size seen in ash2S112411 adults is rescued by expression of ash2Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4nub-AC-62.

Expression of ash2Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4Bx-MS1096 rescues the partial ventralisation of the dorsal wing margin bristles which is seen in ash2S112411 mutants.
