Expression of Gα49BdsRNA.Scer\UAS under the control of Scer\GAL4Orco.T:Hsim\VP16 does not reduce the odour response of the ab1A or ab2A olfactory receptor neurons (tested with ethyl acetate) or of the ab3A ORN (tested with methyl butyrate).

Expression of Gα49BdsRNA.Scer\UAS under the control of Scer\GAL4Gr21a.9.323 results in a significant decrease in the response of the ab1C ORN to CO[[2]], across a wide response range.

Expression of Gα49BdsRNA.Scer\UAS under the control of Scer\GAL4TrpA1.PR impairs thermotactic selection of 18 over 24[o]C.

Expression of Galpha49BdsRNA.Scer\UAS under the control of either Scer\GAL4c309, Scer\GAL4247 (both drive expression in all lobes of the mushroom bodies) or Scer\GAL4Tab2-201Y (in the γ-lobes) leaves spontaneous odor intensity discrimination and avoidance of individual odors unchanged.

Expression of Galpha49BdsRNA.Scer\UAS under the control of either Scer\GAL4c309, Scer\GAL4247 (both drive expression in all lobes of the mushroom bodies) or Scer\GAL4Tab2-201Y (in the γ-lobes) severely disrupts spontaneous odor identity discrimination and mildly disrupts conditioned odor identity discrimination but does not disrupt conditioned odor intensity discrimination.

Spontaneous odor identity discrimination is significantly disrupted in transgenic Scer\GAL4c309/+; Galpha49BdsRNA.Scer\UAS, Scer\GAL80ts.αTub84B/+ flies at the restrictive (30[o]C) but not the permissive temperature.

The amplitude of light induced quantum bumps in the photoreceptors of flies expressing Gα49BdsRNA.Scer\UAS under the control of Scer\GAL4GMR.PF is reduced compared to wild type (when ATP is present in the intracellular solution during recordings).

When Gα49BdsRNA.Scer\UAS is driven by Scer\GAL4Or83b.T:Hsim\VP16, olfactory behaviour is disrupted. Mutant animals appear insensitive to isoamyl acetate compared to wild-type. Response to benzaldehyde at 10-5M is normal, but at 10-4M benzaldehyde induces avoidance behaviour in mutant flies compared to attraction in controls. Responses to crushed apples or Drosophila food are normal. Mutant flies also exhibit an impaired ability to chemotax into traps containing yeast extract.
