Expression of mthScer\UAS.cSa under the control of Scer\GAL4GMR.PU has no effect on eye morphology.
When mthScer\UAS.cSa is driven pre-synaptically by Scer\GAL4D42, the evoked EJP amplitude is increased to about 157% of their normal value. When driven post-synaptically by Scer\GAL4how-24B no effects are seen on evoked EJP amplitudes. Neither driver has a significant effect on mEJP amplitudes or frequency. When driven by Scer\GAL4hs.PB in the absence of heat shock, evoked EJP amplitudes are increased to about 124% of controls, and thirty to sixty minutes after a single heat shock, EJP amplitudes are increased to about 154% of their original value.
mthUAS.cSa/Scer\GAL4hs.PB rescues mth1/mthΔ6
Scer\GAL4how-24B/mthUAS.cSa fails to rescue mth1/mthΔ6
In the absence of heat shock, EJP amplitudes are slightly but significantly rescued compared to mth1/mthΔ6 mutants. After heat shock the EJP amplitudes are statistically indistinguishable from wild-type. The ultrastructural defects in synaptic size and vesicle docking and clustering are also rescued. When mthScer\UAS.cSa is driven by Scer\GAL4how-24B post-synaptically no effect is seen on EJP amplitude or frequency. When driven pre-synaptically by Scer\GAL4D42 however, EJP amplitudes are increased from about 60% to about 100% of control amplitudes while mEJP amplitudes and frequency are not affected. This expression restores evoked release at mutant neuromuscular junctions, since the resulting EJP amplitudes are significantly different from mutant controls but indistinguishable from controls.
