The median lifespan of Sir217 flies (mutant line generated by outcrossing the mutation into the Canton S background) responds in the same way as that of Canton S controls to a range of diets (from dietary restriction to fully-fed conditions).

Sir217 heterozygotes do not display abnormal longevity.

Heterozygous males do not have ectopic sex combs on the T2 or T3 legs.

Sir217/+ suppresses the defect in oocyte commitment (high frequency of 'two-oocyte phenotype' in region 3 of the germarium) that is seen in polo1/poloS025604 germaria.

The frequency of ectopic sex combs seen in Pc3/+ males is increased by Sir217; in Sir217/+ ; Pc3/+ double heterozygous males, 94% of T2 legs and 63% of T3 legs have sex combs. The frequency of ectopic sex combs seen in Pcl11/+ males is increased by Sir217; in Sir217/+ ; Pcl11/+ double heterozygous males, 71% of T2 legs and 38% of T3 legs have sex combs. Eye colour: Sir217/+ moderately, though consistently, impairs the silencing of the heterozygous wUbxPRE.2.2 transgene, resulting in a slight increase in eye pigmentation.

The increased average lifespan of Rpd3def24 homozygous flies is suppressed by Sir217/+.

When Scer\GAL4elav-C155>Hsap\HTTQ93.ex1p.Scer\UAS-expressing animals are also heterozygous for Sir217, the extent of neuronal loss is reduced and show improved motor function compared with siblings wild type Sir2. However, homozygous Sir217 enhances the neuronal loss phenotype and exacerbates the defects in motor function.

A heterozygous Sir217 background markedly increases the survival rate of flies neuronally-expressing Hsap\HTTQ93.ex1p.Scer\UAS flies (from <10% to nearer 35% survival to eclosion). In addition, the level of neurodegeneration, as measured by the ratio of rhabdomeres per ommatidia, is also lowered in a Sir217 heterozygous background.
