Expression of foxoScer\UAS.cKa under the control of Scer\GAL4Act5C.PI early in larval development results in complete developmental arrest of the larvae, which remain in the first instar for up to 7 days. The feeding behaviour of these larvae is different from controls; the larvae expressing foxoScer\UAS.cKa under the control of Scer\GAL4Act5C.PI show a 3-4 fold increase in wondering at 48 and 72 hours after egg laying compared to control larvae. Expression of foxoScer\UAS.cKa under the control of Scer\GAL4hs.PB early in larval development using heat shock treatment for 10 minutes every 24 hours inhibits larval growth. This developmental arrest is reversible. Expression of foxoScer\UAS.cKa under the control of Scer\GAL4hs.PB in the third larval instar using heat shock at 37oC for 4 hours results in significant lethality. The rare escapers that do survive to adulthood are smaller than control flies. Expression of foxoScer\UAS.cKa under the control of Scer\GAL4hs.PB at 29oC results in flies which are approximately half the weight of control flies. The wing area is reduced nearly one third, due to a reduction in both cell size and cell number. Eyes show reductions in both ommatidia number and ommatidia area (reflecting cell number and cell size respectively). Expression of foxoScer\UAS.cKa under the control of Scer\GAL4GMR.PF results in eyes that are smaller than normal, and they lack many ommatidia and nearly all of the mechanosensory bristles. The remaining ommatidia are arranged in the typical hexagonal array and all of the normal photoreceptor cells are present. Expression of foxoScer\UAS.cKa under the control of Scer\GAL4GMR.PF results in a significant reduction in the area of the ommatidia compared to control flies.

Co-expression of foxoScer\UAS.cKa and Pi3K92ED954A.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4GMR.PF almost completely obliterates the eyes. Co-expression of Akt1Scer\UAS.cSa or Pi3K92EScer\UAS.T:Hsap\MYC almost completely rescues the eye phenotype caused by expression of foxoScer\UAS.cKa under the control of Scer\GAL4GMR.PF; the ommatidia and nearly all of the mechanosensory bristles are restored. Co-expression of EgfrScer\UAS.cBa and foxoScer\UAS.cKa under the control of Scer\GAL4GMR.PF does not rescue the eye phenotype seen in animals expressing foxoScer\UAS.cKa alone under the control of Scer\GAL4GMR.PF (ommatidia and bristles are still missing) and the general disorganisation of the ommatidia seen in animals expressing EgfrScer\UAS.cBa alone under the control of Scer\GAL4GMR.PF is also unaffected in the double mutant flies. Co-expression of Ras64BVal14.Scer\UAS is sufficient to restore many of the ommatidia and mechanosensory bristles that are lost in the eyes of animals expressing foxoScer\UAS.cKa.
