Description

Imprecise excision of P{PZ}cup04506 has caused a deletion of a 1466 bp. The remaining cup coding region allele is predicted to encode a truncated protein lacking the first 347 amino acids.

Detailed Description

cup20/cupΔ212 or cupΔ212/cup8 third instar larvae have significant increases in small extra satellite boutons (for example, budding from parent type 1b boutons) at the (muscle 4) neuromuscular junction (NMJ), compared to wild type or heterozygote controls; Df(2L)BSC7/cupΔ212 show significant increases compared to wild type but not Df(2L)BSC7/+ heterozygotes; cupΔ212/+ heterozygotes show significant increases in boutons compared to wild type.

cupΔ212/Df(2L)BSC7 third instar larvae do not show significant changes in evoked excitatory postsynaptic potential (EPSP) amplitudes (recordings from muscle 6/7 NMJs), but miniature EPSP (mEPSP) frequency (but not amplitude) is significantly increased compared to controls.

cupΔ212 homozygous females lay fragile eggs. The cytoskeletal polarity of cupΔ212 homozygous stage 9 oocytes appears normal.

cupΔ212/+;Df(3L)BSC113/+, cupΔ212/+;EndoAΔ4/+, cupΔ212/+;Dadj1E4/+ or cupΔ212/Df(2L)BSC302 transheterozygotes show significant increases in satellite boutons at the NMJ of third instar larvae compared to either single heterozygote. cupΔ212/Df(2R)X58-7 or cupΔ212/+;Df(3R)3450/+ transheterozygotes do not show significant increases in satellite boutons at the NMJ of third instar larvae compared to either single heterozygote.

cupΔ212/+ has a very weak effect on the penetrance of the mutant dorsal appendage phenotype seen in eggs laid by BicC4/+ females, increasing it from 5% to 9% in the double heterozygous flies.

Comments

Expression of cupScer\UAS.cMa driven by Scer\GAL4elav.PU significantly rescues the satellite bouton phenotype seen at the NMJ of Df(2L)BSC7/cupΔ212 or Df(2L)BSC7/cup20 third instar larvae; expression driven by Scer\GAL4how-24B fails to rescue the phenotype in Df(2L)BSC7/cup20 larvae.
