Coding region for CrebB is corrected to wild type sequence by replacement of first 139bp of the mutated CrebB cassette in CrebBmut.UAS with wild type sequences from the CrebB isoform b. Expression is governed by UAS regulatory sequences.
Overexpression of CrebB-17Acor.Scer\UAS driven by Scer\GAL4elav.PLu causes a mild, but significant, increase in synaptic bouton numbers at larval neuromuscular junctions.
Heat shock induced expression of CrebB-17Acor.Scer\UAS causes a slow lethality. Four days after 30 min heat shock at 37oC over 50% of the flies were dead, with lethality reaching 99% by day 7. Without heat shock, though in combination Scer\GAL4hs.PB with which nevertheless increases expression of CrebB-17Acor.Scer\UAS, the positive effect of 5x spaced training in the olfactory classical conditioning LTM assay (tested at 4 days) is abolished. Thus expression depresses learning in this assay.
