Description

UAS sequences drive expression of a pair of complementary 403bp fragments of Rab11, resulting in a construct which should produce Rab11 dsRNA.

Expression of Rab11dsRNA.UAS.WIZ under the combined control of Scer\GAL4Ugt36A1.PK and Gal80[ts] (for the temporal control of expression) leads to slit diagram localization defects (localized loss of slit diaphragms and shortened lines of slit diaphragm protein on the surface upon 2d expression; and extensive loss of slit diaphragm protein from the membrane upon 4d expression).

Expression of Rab11dsRNA.Scer\UAS.WIZ in somatic clones under the control of Scer\GAL4Act5C.PI results in cells with short, missing or malformed trichomes in pupal wings.

Flies expressing Rab11dsRNA.Scer\UAS.WIZ under the control of Scer\GAL4ninaE.PT which are raised at 18[o]C prior to eclosion and then raised under 12 hour light/12 hour dark conditions show a reduction in the amplitude of the electroretinogram at 3 weeks of age. This retinal degeneration defect is not seen if the flies are raised in constant darkness after eclosion.

Expression of Rab11dsRNA.Scer\UAS.WIZ in the eye, under the control of Scer\GAL4GMR.PF results in R-cells failing to maintain their apical localization. Instead of forming a tightly associated R-cell cluster in the apical region, as observed in wild-type, R-cell clusters in Rab11dsRNA.Scer\UAS.WIZ mutants extend in a long stretch with the distribution of R-cell nuclei along the entire apical-basal axis.

Expression of Rab11dsRNA.Scer\UAS.WIZ under the control of Scer\GAL4hs.PB results in small rhabdomeres.
