Contains 4.5kb of U2af50 upstream genomic sequence, followed by the 216bp U2af50 5' UTR with an improved Cavener translation initiation sequence, followed by the tripeptide MAS and the full female-specific tra open reading frame, followed by the 77bp U2af50 3' UTR and adjacent 1.5kb downstream genomic sequence.
The XY germline degenerates in XY animals expressing traF.U2af50.
In mosaic XX/XO animals (gynanders) which are carrying traF.U2af50, where all the somatic cells of both gonads (those cells inside of the epithelial sheath that surrounds each ovariole) are XO, then the XO somatic cells support normal oogenesis of XX germ cells, resulting in morphologically wild-type eggs. 33% of these gynanders lay their eggs and all those that lay eggs are fertile.
In mosaic XX/XO animals (gynanders) which are carrying traF.U2af50, XO germ cells form tumorous egg chambers, both when the somatic cells of the gonad (those cells inside of the epithelial sheath that surrounds each ovariole) are XO and also when the somatic cells of the gonad are XX.

traF.U2af50 feminizes Sxlf7,M1/SxlM1,fΔ33 mutants without increasing their size. Sxlf7,M1/SxlM1,fΔ33 ; traF.U2af50/+ females have large ovaries full of mature eggs, but they fail to lay.
Phf7ΔN2 significantly rescues germ cell number and organisation in XY animals expressing traF.U2af50.
