UAS regulatory sequences drive expression of a 585bp cDNA fragment (nucleotides 571-1156) cloned in one orientation and then in the reverse orientation to generate a hairpin with a small intronic sequence in between.

Expression of 5-HT1BIR.Scer\UAS under the control of Scer\GAL4tim.PE does not affect either the total amount of sleep or sleep consolidation, measured as the average length of sleep bouts.

Flies in which 5-HT1BIR.Scer\UAS is expressed under the control of Scer\GAL4tim.PE exhibit significantly increased light-induced phase shifts at low light intensity (250 lux) as compared to wild-type. In addition, 5-HTP treatment does not inhibit a strong light response in these flies.

Expression of 5-HT1BIR.Scer\UAS, under the control of Scer\GAL45-HT1B in flies that have been subjected to light pulses of different intensities results in significantly increased phase shifts as compared to wild-type flies. These flies are also immune to 5-HTP treatment.

Flies in which 5-HT1BIR.Scer\UAS is expressed under the control of Scer\GAL4tim.PE show significantly increased light-induced phase shifts at low light intensity as compared to control flies. In addition, 5-HTP treatment does not inhibit the light response of these flies.

Expression of 5-HT1BIR.Scer\UAS specifically in cells that normally express 5-HT1B (i.e. under the control of Scer\GAL45-HT1B) results in circadian periods significantly longer than those of controls. The same effect is observed when 5-HT1BIR.Scer\UAS is expressed under the control of Scer\GAL4elav-C155.

Reducing 5-HT1B expression, through induction of 5-HT1BIR.Scer\UAS, under the control of Scer\GAL4tim.PE, eliminates the strong behavioural rhythms in constant bright light observed in cryb heterozygous flies overexpressing 5-HT1BScer\UAS.cYa under the control of Scer\GAL4tim.PE.
