Fewer autolysosomes are formed in the fat body cells of Rack11.8 mutant larvae during starvation compared to controls, and the ones that are seen are small. The cytoplasmic area covered by autophagosomes and autolysosomes is also decreased. Glycogen stores are also strongly decreased in Rack11.8 mutants, with an 11.8 fold reduction in glycogen particle area compared to controls.

75% of Rack11.8 mutants show larval lethality and of those that survive to pupariate, 85% die before adult eclosion.

When females carrying Rack11.8 germ-line clones are mated to wild-type males, the females display a dramatically reduced rate of egg lay and aberrantly small ovaries. The produced eggs are reduced in size by an average of 30% compared to wild-type eggs. However, these eggs are competent to be fertilized, although fertilized embryos arrest very early in embryogenesis.

Some Rack11.8/Df(2L)RACK1-mts mutant germaria contain numbers of germline stem cells (GSCs) that are within the normal range, while others are completely devoid of GSCs. Ovarioles that contain no GSCs contain more mature egg chambers. Some Rack11.8/Df(2L)RACK1-mts egg chambers contain fewer germ cells per chamber and a large portion of chambers contain either four or eight nuclei. Additionally, the number of ring canals does not always correspond to the number of nuclei present.

Rack11.8/Df(2L)RACK1-mts, Rack11.8/Rack1EY00128 and Rack11.8/Rack1EE escapers display a range of phenotypes including sluggish movement, difficulty in exiting the pupal case, cuticle that does not harden, and wings that are blistered, abnormally small, or that fail to unfold. Escaper females are incapable of laying eggs and the size of the ovary is reduced compared to wild type but overall body size is unaffected.
