UASt regulatory sequences drive expression of an inverted repeat.
No adult flies expressing P{GD1710} under the control of Scer\GAL4Act5C.PU in a GgaΔ background are recovered. This illustrates that the lethality associated with expression of P{GD1710} under the control of Scer\GAL4Act5C.PU must be an off-target effect (not due to an effect on Gga), since the Gga sequences targeted by the P{GD1710} construct are completely deleted in the GgaΔ allele.
When GgaGD1710 is expressed under the control of Scer\GAL4Act5C.PU, fewer flies than expected survive to adulthood.
Flies expressing GgaGD1710 ubiquitously under the control of Scer\GAL4αTub84B.PL develop normally, with no gross morphological changes. There are no differences in fertility in both males and females, neither in rate, number or overall development of progeny. There are no qualitative morphological differences in the development of sperm.
Adults expressing GgaGD1710 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
