Expression of one copy of Gem3ΔN.Scer\UAS driven by Scer\GAL4Mef2.PR has no effect on both motor function and survival throughout adulthood; expression of two copies of Gem3ΔN.Scer\UAS results in the majority of flies being flightless (5 days old) and flight ability progressively declines (none fly by day 15) and there is a progressive drop in adult survival.

Flies expressing Gem3ΔN.Scer\UAS (co-expressed with Dcr-2Scer\UAS.cDa) under the control of Scer\GAL4Mef2.PR show an age-dependent reduction in climbing ability compared to controls. The majority of flies are flightless at 5 days post-eclosion.

Flies co-expressing Gem3Scer\UAS.cCa, Gem3GD17221 and Gem3ΔN.Scer\UAS under the control of Scer\GAL4Mef2.PR are flight defective.

Animals expressing Gem3ΔN.Scer\UAS under the control of Scer\GAL4how-24B show several defects at metamorphosis; the flies are "headless" as a results of failed head eversion and the wings are shortened. The pupae are elongated compared to wild type. The abdomen remains elongated (as in the larval stage) during pupal development in these animals, although its shows normal segmentation as shown by the complete bristle pattern on the tergites. The leg discs evert but the legs are not fully elongated.

Eclosed flies expressing Gem3ΔN.Scer\UAS under the control of Scer\GAL4Mef2.PR have a reduced ability to fly, although they can walk and jump normally. The indirect flight muscles in these animals are shrunken, resulting in increased spacing, and breakages and large tears can be seen between the muscle fibres.

Adult flies bearing Df(1)ED6716 and also expressing Gem3ΔN.Scer\UAS under the control of Scer\GAL4Mef2.PR display rapid age-progressive decrease in viability as well as flight ability relative to controls.

Expression of Gem3ΔN.Scer\UAS together with GlosGD17786 under the Scer\GAL4Mef2.PR driver are adult lethal and simultaneous expression of Gem3ΔN.Scer\UAS and any of the following: GlosGD17786, rigNIG.11171R, rigScer\UAS.T:Avic\GFP-ECFP, Gem2Scer\UAS.cBa, SmndsRNA.C.Scer\UAS.WIZ or SmnScer\UAS.T:Zzzz\FLAG leads to strongly reduced mobility in third instar larvae and more elongated shape of the puparium.

Scer\GAL4elav-C155-driven co-expression of Gem3ΔN.Scer\UAS and GlosGD17786, Gem2Scer\UAS.cBa, SmndsRNA.C.Scer\UAS.WIZ or SmnScer\UAS.T:Zzzz\FLAG reduced adult viability and Gem3ΔN.Scer\UAS;rigScer\UAS.T:Avic\GFP-ECFP flies are completely lethal.

Flies expressing simultaneously VletGD5216 and Gem3ΔN.Scer\UAS under the control of Scer\GAL4Mef2.PR display significantly reduced survival rate compared to controls.

Df(3R)Exel6178 adult heterozygotes expressing Gem3ΔN.Scer\UAS under the control of Scer\GAL4Mef2.PR display age-progressive loss of flight ability.

No decline in adult flight ability compared to controls is observed when wmdKK100974 and Gem3ΔN.Scer\UAS are co-expressed under the Scer\GAL4Mef2.PR driver.

Expression of Tgs1GD14932 together with Gem3ΔN.Scer\UAS under the control of Scer\GAL4Mef2.PR leads to age-progressive flight defects.

Co-expression of SmnΔ6.Scer\UAS.T:Avic\GFP and Gem3ΔN.Scer\UAS driven by Scer\GAL4Mef2.PR results in flightless flies and a significant drop in adult viability throughout adulthood. Co-expression of SmnScer\UAS.T:Zzzz\FLAG and Gem3ΔN.Scer\UAS driven by Scer\GAL4Mef2.PR results in partial lethality, with the few escapers exhibiting severe flight defects. Co-expression of Gem3ΔN.Scer\UAS and SmndsRNA.C.Scer\UAS.WIZ driven by Scer\GAL4Mef2.PR results in flight defects and reduces adult viability, compared to controls.

Co-expression of Gem3ΔN.Scer\UAS and Gem2ΔC.Scer\UAS driven by Scer\GAL4Mef2.PR results in progressive age-dependent flight defects (no significant effect on survival with age progression). Co-expression of Gem3ΔN.Scer\UAS and one copy of Gem2NIG.10419R driven by Scer\GAL4Mef2.PR results in flight defects and partial lethality; co-expression of Gem3ΔN.Scer\UAS and two copies of Gem2NIG.10419R driven by Scer\GAL4Mef2.PR results in lethality.
