Sense-antisense transcription (driven by two convergent arrays of UAS regulatory sequences) of a 686bp fragment of vtd exon 4 (nucleotides 1152-1837, coordinates relative to GenBank accession AF109926) results in double stranded RNA (dsRNA) expression.

Expression of vtddsRNA.Sym.Scer\UAS under the control of Scer\GAL4Act5C.PI results in reduced survival or both male and female adults, with the males being more strongly affected. The lethality is dramatically enhanced if the animals are also heterozygous for vtd80Fh-1.

Expression of P{Sym-UAS-Rad21} (which carries both vtdSym.Scer\UAS and vtda.Sym.Scer\UAS) under the control of Scer\GAL4Act5C.PI results in reduced survival or both male and female adults, with the males being more strongly affected. The lethality is dramatically enhanced if the animals are also heterozygous for vtd80Fh-1.

Expression of vtddsRNA.Sym.Scer\UAS under the control of Scer\GAL4Act5C.PI can result in pupal lethality, depending on the particular P{Sym-UAS-Rad21} insertion line used. No increase in precocious sister chromatid separation in third larval instar brain squashes is seen in these animals.
