Co-expression of StimScer\UAS.cAa and olf186-FScer\UAS.cZa driven by Scer\GAL4Ilp2.PR completely suppresses the wing posture, flight and flight physiology defects of Itp-r83Aka1091/Itp-r83Aug3 flies.

Simultaneous expression of StimScer\UAS.cAa and olf186-FScer\UAS.cZa driven by Scer\GAL4Ddc.PL in aminergic neurons results in pupal lethality.

Expression of StimScer\UAS.cAa driven by either Scer\GAL4Ilp2.PR or Scer\GAL4Ddc.PL suppresses the altered wing posture of Itp-r83Aka1091/Itp-r83Aug3 flies as well as the spontaneous hyperactivity of the dorsal longitudinal muscles. The flight ability and sustained air-puff-induced rhythmic flight patterns are also restored in these flies.

Pan-neuronal expression of StimScer\UAS.cAa and olf186-FScer\UAS.cZa driven by Scer\GAL4elav-C155 in Itp-r83Aka1091/Itp-r83Aug3 flies suppresses the abnormal wing-posture to varying extents but does not fully restore flight ability.

Overexpressing StimScer\UAS.cAa with or without olf186-FScer\UAS.cZa driven by Scer\GAL4Ilp2.PR in Itp-r83Aka1091/Itp-r83Aug3 mutant neurons restores global intracellular Ca[2+] homeostasis.

Scer\GAL4nSyb.PS-driven expression of StimScer\UAS.cAa does not rescue small larval body size or the delay in larval development characteristic for Stimko/Stimko but it does partially rescue their lethality - as some adults manage to eclose. Similarly, the lethality but not the developmental delay is partially rescued by Scer\GAL4ple.PF- or Scer\GAL4ple.TH-A-driven expression and the lethality is almost completely rescued when StimScer\UAS.cAa is driven by Scer\GAL4Act5C.PI.

Co-expression of StimScer\UAS.cAa fully restores the developmental delay characteristic for flies expressing StimsgRNA.dual.snRNA:U6:96Ab together with Spyo\Cas9Scer\UAS.T:nls-Ude under the control of Scer\GAL4elav-C155 (to induce tissue-specific gene knock-out) but their increased lethality is only partially rescued.
