Detailed Description

Homozygous adults start to show defects in climbing ability and their ability to fly at 20 days of age, and then die within 30 days of eclosion.

Homozygous males are sterile. No mature sperm are seen in the vas deferens. The early stages of spermatogenesis appear normal but the mutant males show defects in spermatid individualization; the morphology of the cystic bulges and waste bags is abnormal. The nebenkern are less condensed than normal in the mutant spermatids.

Mutant flies show a similar survival rate as control flies after feeding with 20mM paraquat.

Mutant eye discs show a similar level of apoptosis following incubation with an ionophore as do control eye discs.

Homozygous and porinA2/Df(2L)BSC143 adults show defects in mitochondrial morphology in the indirect flight muscles, with elongated tube-like mitochondria being seen between the muscle fibers.

porinA2/+ or porinA2/porinA2 does not suppress the small eye phenotype seen in animals expressing debclScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PF. The increased apoptosis seen in the eye discs of these animals is also not suppressed by either porinA2/+ or porinA2/porinA2.

The defect in mitochondrial morphology in the indirect flight muscles which is seen in porinA2 homozygotes is suppressed by expression of either Drp1Scer\UAS.T:Ivir\HA1 or MarfGD11094 under the control of Scer\GAL4Mef2.PR.

The defects in flight ability that are seen in porinA2 flies 20 days after eclosion are rescued by expression of Drp1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4Mef2.PR.

Comments

Expression of porinScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4tub.PU rescues the defects in climbing and flight ability that are seen in porinA2 flies 20 days after eclosion.

Expression of porinScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4tub.PU rescues the sterility of porinA2 homozygous males.

Expression of porinScer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4tub.PU significantly rescues the defects in mitochondrial morphology in the indirect flight muscles that are seen in porinA2 homozygotes.
