Inhibition (using shi1.UAS at 32[o]C) of Scer\GAL4GMR38E07 cells in adults during testing blocks the increase in aversive olfactory 24 h memory observed when the recall environment better matches the training environment (presence of copper grid), compared to controls.
Acute inhibition (using shi1.UAS at 30[o]C) of a subset of Scer\GAL4GMR38E07 cells (expression restricted using Scer\GAL80Mef2.mb247) in adults significantly reduces electric shock avoidance, particularly at higher voltages, and avoidance of higher temperatures, compared to controls, with no effect on locomotion activity index or odor avoidance; inhibition of this subset during testing alone, or training alone, has no effect on short-term aversive, olfactory memory, compared to controls.
Inhibition (using Hsap\KCNJ2UAS.EGFP) of Scer\GAL4GMR38E07 cells, in combination with a Gal80[ts] transgene to begin expression three days prior to experiments, in adults significantly reduces electric shock avoidance, compared to controls, with no effect on locomotion activity index.
Optogenetic activation (using Crei\ChR1::Vcar\ChR1::Vcar\ChR2ReaChR.UAS.Citrine with (orange) light) of Scer\GAL4GMR38E07 cells in one chamber of a T-maze results in a significant increase in avoidance of that chamber, compared to controls, with no effect on locomotion activity index.
Thermogenetic activation (using TrpA1UAS.(B).cKa at 26[o]C) of a subset of Scer\GAL4GMR38E07 cells (expression restricted using Scer\GAL80Mef2.mb247) in adults, in combination with a paired odor, results in a significant increase in aversive olfactory memory, compared to controls, with no effect on locomotion activity index or odor avoidance.
