Adulthood-only expression of Atg12HMS01153 under the control of Scer\GAL4esg.PU leads to a significant increase in the number of mitotic cells (phospho-H3 staining) in the adult midgut, as compared to controls. (The temporal regulation of expression is dependent on Gal80[ts] and induced by temperature shift to 29[o]C for 5-6 days during adulthood).

Adulthood-only expression of Atg12HMS01153 under the combined control of Scer\GAL4Myo31DF-NP0001 and Gal80[ts] does not significantly affect the posterior midgut cell size, as compared to controls. Adulthood-only expression under the combined control of Scer\GAL4tub.PU and Gal80[ts] leads to the failure to induce autophagy (assessed by the lysosome and autophagosome marker, Lysotracker) in the adult fat body in response to starvation, as compared to controls.

The clonal expression of Atg12HMS01153 under the control of Scer\GAL4Act.PU results in a significant increase in the transverse cell area and a significant increase in relative mitochondrial content of midgut enterocytes during pupariation, as compared to controls.

Expression of Atg12HMS01153 under the control of Scer\GAL4Mef2.PR is able to significantly suppress the formation of autophagosomes in the muscles of chloroquine-treated larvae starved on low-nutrient food for 6 hours.
