UAS regulatory sequences positioned at either end of the coding region drive expression of a 654bp RpS6 fragment in both sense and antisense orientation, resulting in the generation of dsRNA.

Expression of RpS6dsRNA.Sym.Scer\UAS under the control of Scer\GAL4GMR.PU results in a smaller eye than normal which has a glassy appearance and necrotic patches.

Expression of RpS6dsRNA.Sym.Scer\UAS under the control of Scer\GAL4ey.PU does not result in an obvious mutant eye phenotype.

Expression of RpS6dsRNA.Sym.Scer\UAS under the control of Scer\GAL4amn-c651 results in a reduction in the size of the prothoracic gland in larvae compared with controls at the equivalent time point of 5 days after egg deposition (AED). In addition, these animals show an extreme developmental delay compared to wild type: at day 5 AED the larvae are smaller than control larvae, and they continue to grow and feed beyond day 10 to become giant larvae that fail to pupate. Addition of 20-hydroxyecdysone to the food results in a variable restoration of pupariation, but the final steps of metamorphosis, including the formation of adult structures, are not rescued.

Animals in which expression of RpS6dsRNA.Sym.Scer\UAS under the control of Scer\GAL4amn-c651 is delayed until the late second larval instar (using the temperature sensitive Scer\GAL80ts.αTub84B allele) are able to undergo pupation and eclose as large adults. These animals show an increase in eye size and in wing size compared to controls. If the animals are raised in the presence of 20-hydroxyecdysone, the body size of the adults is restored to normal.
