Expression of GgadsRNA.Scer\UAS.2 under the control of Scer\GAL4GMR.PU results in age-dependent retinal degeneration.
No adult flies expressing P{UAS-Gga.RNAi.2} under the control of Scer\GAL4Act5C.PU in a GgaΔ background are recovered. This illustrates that the lethality associated with expression of P{UAS-Gga.RNAi.2} under the control of Scer\GAL4Act5C.PU must be an off-target effect (not due to an effect on Gga), since the Gga sequences targeted by the P{UAS-Gga.RNAi.2} construct are completely deleted in the GgaΔ allele.
When GgadsRNA.Scer\UAS.2 is expressed under the control of any one of Scer\GAL4Act5C.PU, Scer\GAL4mat.αTub67C.T:Hsim\VP16 or Scer\GAL4da.G32, no adults emerge. The lethality occurs during early pupation with all three drivers.
